RESUMEN

Chronic inflammation promotes the development of pancreatic ductal adenocarcinoma (PDAC) and PDAC-related inflammatory tumor microenvironment facilitates tumor growth and metastasis. Thus, we aimed to study the association between inflammatory response and prognosis in patients with PDAC. We conducted the whole transcriptomic sequencing using tissue samples collected from patients diagnosed with PDAC (n = 106) recruited from Shandong Cancer Hospital. We first constructed a prognostic signature using 15 inflammation-related genes in The Cancer Genome Atlas (TCGA) cohort (n = 177) and further validated it in an independent International Cancer Genome Consortium (ICGC) cohort (n = 90) and our in-house cohort. PDAC patients with a higher risk score had poorer overall survival (OS) (P < 0.001; HR, 3.02; 95% CI, 1.94-4.70). The association between the prognostic signature and OS remained significant in the multivariable Cox regression adjusting for age, sex, alcohol exposure, diabetes, and stage (P < 0.001; HR, 2.91; 95% CI, 1.73-4.89). This gene signature also robustly predicted prognosis in the ICGC cohort (P = 0.01; HR, 1.94; 95% CI, 1.14-3.30) and our cohort (P < 0.001; HR, 2.40; 95% CI, 1.45-3.97). Immune subtype C3 (inflammatory) was enriched and CD8+ T cells were higher in patients with a lower risk score (P < 0.05). Furthermore, PDAC patients with higher risk scores were more sensitive to chemotherapy, immunotherapy, and PARP inhibitors (P < 0.05). In sum, we identified a novel gene signature that was associated with inflammatory response for risk stratification, prognosis prediction, and therapy guidance in PDAC patients. Future studies are warranted to validate the clinical utility of the signature.

Sibling relationships are most individuals' longest lasting relationships, but their development remains understudied. Using a within-family, accelerated longitudinal design with data from mothers, fathers, and two siblings from 201 predominately White, working-, and middle-class families, we charted the development of sibling intimacy and conflict from age 7 to age 30. We also examined structural characteristics (sibling sex, sex constellation, age spacing, birth order) and both person mean (between-person) and time-varying (within-person) links between (a) feminine-typed, expressive personality and (b) maternal and paternal warmth and conflict and sibling intimacy and conflict, respectively, and tested whether sibling age moderated these linkages. Overall cubic changes suggested that intimacy declined from middle childhood through early adolescence and increased through the mid-20s and leveled off, whereas conflict was stable in middle childhood, declined across adolescence, and leveled off in young adulthood, with patterns that varied by sibling sex, dyad sex constellation, and birth order. Furthermore, expressivity was related to both sibling intimacy (positively) and conflict (negatively) at between- and within-person levels; maternal warmth was positively related to sibling intimacy and maternal conflict was positively related to sibling conflict at both between- and within-person levels; and father warmth and conflict were linked to sibling intimacy and conflict, respectively, at the within-person level. Finally, age moderated expressivity effects such that these disappeared in young adulthood and moderated the between-person father warmth effect such that this effect emerged in young adulthood and increased through age 30. An instant p-doping strategy employing 4-tert-butyl-2-chloropyridine and tert-butyl peroxybenzoate for the spiro-OMeTAD hole-transport layer (HTL) in perovskite solar cells (PSCs) is proposed to replace the conventional 4-tert-butylpyridine-doped HTL. The novel doping process eliminates the formation of pores in the HTL. Meanwhile, a 21.4% efficiency is achieved on the corresponding absolute methylammonium-free PSCs with significantly improved thermal and moisture stability.

Microplastics (MPs) are a serious threat to the living environment of aquatic organisms. However, there are fewer studies on the toxicity of microplastics to freshwater organisms. This study aimed to establish a polystyrene microplastics (PS-MPs) model by feeding carp (Cyprinus carpio) PS-MP (1000 ng/L) particles 8 µm in size. HE staining revealed a mass of inflammatory cells infiltrated in the carp hepatopancreas. The activities of alkaline phosphatase (AKP), aspartate transaminase (AST), lactate dehydrogenase (LDH), and alanine transaminase (ALT) were strengthened considerably, suggesting that PS-MPs cause injury to the hepatopancreas of carp. Real-Time polymerase chain reaction and western blotting results indicated increased levels of glucose-regulated protein 78 (GRP78), (PKR)-like ER kinase (PERK), eukaryotic translation initiation Factor 2α (EIF2α) and activating transcription Factor 4 (ATF4) genes and increased levels of inflammatory factors downstream of endoplasmic reticulum stress (ERs) thioredoxin-interacting protein (TXNIP), NOD-like receptor protein 3 (NLRP3), interleukin-18 (IL-18), interleukin-1ß (IL-1ß), and caspase 1. Increased expression of microtubule-associated protein-2 (LC3II), autophagy-related 5 (ATG5) and autophagy-related 12 (ATG12) genes revealed that PS-MPs promoted autophagy in carp hepatocytes. The enhanced expression of the Caspase 12, Caspase 3, and Bax genes suggested that PS-MPs led to the apoptosis of carp hepatocytes. These results suggest that PS-MPs result in serious injury to the hepatopancreas of carp. The present study of PS-MPs in freshwater fish from the aspect of endoplasmic reticulum stress was conducted to provide references and suggestions for toxicological studies of PS-MPs in freshwater environments.

The herbicide glyphosate, N-(phosphonomethyl)glycine, has been widely used in the past 40 years, and has had many adverse effects on human health. Here, we constructed a convenient "on-off-on" fluorescent platform for detection of glyphosate via Cu2+ modulated squaraine dye fluorescence quenching. The squaraine dye F-0 exhibited strong fluorescence, which could be quenched by the addition of Cu2+. However, the addition of glyphosate restored the fluorescence intensity of F-0 due to the formation of a Cu2+-glyphosate complex. F-0 was utilized as a fluorescent probe for the quantitative detection of glyphosate, with the lowest detection limit of 13.16 nmol L-1. Furthermore, this method demonstrated high selectivity and anti-interference capabilities. The successful monitoring of glyphosate in real samples was achieved using this detection strategy.

Vascular diseases, a leading cause of death in human, are strongly associated with pathological damage to blood vessels. The selenoprotein (Sel) have been reported to play important roles in vascular disease. However, the role of SelO in vascular disease has not been conclusively investigated. The present experiment was to investigate the regulatory mechanism of the effect of SelO on the permeability of vascular endothelial. The H.E staining, FITC-Dextran staining, Dil-AC-LDL staining and FITC-WGA staining showed that vascular structure was damaged, and intercellular junctions were disrupted with selenium (Se)-deficient. Immunohistochemistry, qPCR and Western blot revealed decreased expression of the adhesion plaque proteins vinculin, talin and paxillin, decreased expression of the vascular connectivity effector molecules connexin, claudin-1 and E-cadherin and increased expression of JAM-A and N-cadherin, as well as decreased expression of the ZO-1 signaling pathways ZO-1, Rock, rhoGEF, cingulin and MLC-2. In a screening of 24 Sel present in mice, SelO showed the most pronounced changes in vascular tissues, and a possible association between SelO and vascular intercellular junction effectors was determined using IBM SPSS Statistics 25. Silencing of SelO, vascular endothelial intercellular junction adverse effects present. The regulatory relationship between SelO and vascular endothelial intercellular junctions was determined. The results showed that Se deficiency lead to increased vascular endothelial permeability and vascular tissue damage by decreasing SelO expression, suggesting a possible role for SelO in regulating vascular endothelial permeability.

Objective:To investigate the detection rate and metastasis rate of delphain lymph node (DLN)in thyroid papillary adenocarcinoma(PTC) and to analyze the risk factors for DLN metastasis. Methods:The clinicopathological data of 200 PTC patients admitted to the from January 2018 to June 2020 were retrospectively analyzed, and the detection of DLN was clearly recorded in the pathological reports of all patients. The number of DLN detected, the number of metastasis, the detection rate and the metastasis rate were counted. The clinicopathological factors that might affect DLN metastasis were analyzed by univariate analysis and multivariate Logistic regression analysis, including gender, age, tumor size and tumor location. Results:DLN was detected in 121 of 200 PTC patients, with a detection rate of 60.50% (121/200). DLN metastasis was found in 46 of the 121 patients with a metastasis rate of 38.02% (46/121).Univariate analysis showed that tumor diameter, multiple foci, capsular invasion, extradandular invasion, lymphatic vascular invasion, lymph node metastasis in central region (excluding DLN), and lateral cervical lymph node metastasis were the risk factors for DLN metastasis of PTC (P<0.05). Gender, age, tumor location, bilateral tumors, Hashimoto's thyroiditis and BRAFV600E mutation were not significantly correlated with DLN metastasis of PTC(P>0.05). The 7 variables with statistically significant differences in univariate analysis were incorporated into Logistic regression model for multivariate analysis, and the results showed that, Tumor diameter ≥1.0 cm, capsule invasion, lymphatic vascular invasion, lymph node metastasis in central region (excluding DLN), and lateral cervical lymph node metastasis were independent risk factors for DLN metastasis of PTC (OR= 3.386-9.186, P<0.05). The sensitivity and specificity of DLN metastasis in predicting central lymph node (excluding DLN) metastasis in PTC patients were 36.79% and 92.55%, respectively, while the sensitivity and specificity of DLN metastasis in predicting lateral cervical lymph node metastasis were 41.03% and 81.37%, respectively.The incidence of central lymph node metastasis (excluding DLN) in DLN-positive patients were was 4.94 times higher than that in DLN-negative patients, and the incidence of lateral neck lymph node metastasis in DLN-positive patients were 2.20 times higher than that in DLN-negative patients. Conclusion:The detection rate and metastasis rate of DLN in PTC patients were higher, DLN metastasis predicts more extensive lymph node metastasis, and DLN metastasis was related to multiple factors,among which tumor diameter ≥ 1.0 cm, capsule invasion, lymphatic vascular infiltration, lymph node metastasis in the central region (excluding DLN), and lateral cervical lymph node metastasis were independent risk factors for DLN metastasis of PTC. Therefore, PTC patients with the above characteristics should actively explore DLN and formulate appropriate surgical strategies.

As a widespread environmental pollutant, microplastics pose a great threat to the tissues and organs of aquatic animals. The carp's muscles are necessary for movement and survival. However, the mechanism of injury of polyethylene microplastics (PE-MPs) to carp muscle remains unclear. Therefore, in this study, PE-MPs with the diameter of 8 µm and the concentration of 1000 ng/L were used to feed carp for 21 days, and polyethylene microplastic treatment groups was established. The results showed that PE-MPs could cause structural abnormalities and disarrangement of muscle fibers, and aggravate oxidative stress in muscles. Exposure to PE-MPs reduced microRNA (miR-21) in muscle tissue, negatively regulated Interleukin-1 Receptor Associated Kinase 4 (IRAK4), activated Nuclear Factor Kappa-B (NF-κB) pathway, induced inflammation, and led to endoplasmic reticulum stress and apoptosis. The present study provides different targets for the prevention of muscle injury induced by polyethylene microplastics.

Grain boundaries of metal halide perovskites contain massive defects that are detrimental to photovoltaics applications. This work demonstrates that inorganic NH4NO3 can selectively passivate the grain boundaries of perovskite films and improve their moisture resistance simultaneously, resulting in enhanced performance and stability of the methylammonium-free perovskite solar cells.

The performance loss caused by encapsulation has been an obstacle to guarantee the excellent power conversion efficiency of perovskite solar cells (PSCs) in practical application. This work revealed that the encapsulation-induced performance loss is highly related to the tensile strains imposed on the functional layers of the device when the PSC is exposed directly to the deformed encapsulant. A barrier strategy is developed by employing a nonadhesive barrier layer to isolate the deformed encapsulant from the PSC functional layer, achieving a strain-free encapsulation of the PSCs. The encapsulated device with a barrier layer effectively reduced the relative performance loss from 21.4% to 5.7% and dramatically improved the stability of the device under double 85 environment conditions. This work provides an effective strategy to mitigate the negative impact of encapsulation on the performance of PSCs as well as insight into the underlying mechanism of the accelerated degradation of PSCs under external strains.

This study explored the collaborative effect on nutrients removal performance and microbial community in solid-phase denitrification based bacteria-algae symbiosis system. Three biodegradable carriers (apple wood, poplar wood and corncob) and two algae species (Chlorella vulgaris and Chlorella pyrenoidosa) were selected in these bacteria-algae symbiosis systems. Results demonstrated that corncob as the carrier exhibited the highest average removal efficiencies of total nitrogen (83.7%-85.1%) and phosphorus removal (38.1%-49.1%) in comparison with apple wood (65.8%-71.5%, 25.5%-32.7%) and poplar wood (42.5%-49.1%, 14.2%-20.7%), which was mainly attributed to the highest organics availability of corncob. The addition of Chlorella acquired approximately 3%-5% of promotion rates for nitrated removal among three biodegradable carriers, but only corncob reactor acquired significant promotions by 3%-11% for phosphorous removal. Metagenomics sequencing analysis further indicated that Proteobacteria was the largest phylum in all wood reactors (77.1%-93.3%) and corncob reactor without Chlorella (85.8%), while Chlorobi became the most dominant phylum instead of Proteobacteria (20.5%-41.3%) in the corncob with addition of Chlorella vulgaris (54.5%) and Chlorella pyrenoidosa (76.3%). Thus, the higher organics availability stimulated the growth of algae, and promoted the performance of bacteria-algae symbiosis system based biodegradable carriers.

BACKGROUND: Immune checkpoint inhibitors (ICIs) have shown efficacy in patients with metastatic urothelial cancer (mUC), however, only a small subset of patients could benefit from ICIs. Identifying predictive biomarkers of ICIs in patients with mUC is clinical meaningful for patient stratification and administration. METHODS: Clinical and transcriptomic data of mUC patients treated with ICIs from mUC cohort (IMvigor210 study) was utilized to explore the predictive biomarkers. LASSO Cox regression was performed to construct a predictive model. The predictive model was trained and tested in the mUC cohort, and then exploratively tested in clear cell renal cell carcinoma (ccRCC) and melanoma cohorts in which patients also received ICIs regimens. RESULTS: The differentially expressed genes (DEGs) in complement and coagulation cascades pathway (CCCP) were mainly enriched in non-responders of ICIs in the mUC cohort. A CCCP risk score was constructed based on the DEGs in CCCP. Patients with a low-risk score were more responsive to ICIs and had better overall survival (OS) than those with a high-risk score in the training set (HR, 0.38; 95%CI, 0.27-0.53, P<0.001) and the test set (HR, 0.34; 95%CI, 0.17-0.71, P=0.003). The association between the CCCP risk score and OS remained significant in the multivariable cox regression by adjusting PD-L1 expression and TMB (P<0.05). In addition, there was no difference for OS in the bladder cancer patients without ICIs (TCGA-BLCA cohort, HR, 0.76, 95%CI, 0.49-1.18, P=0.22), suggesting a predictive but not prognostic effect of the risk score. For the exploratory analysis, consistent results were observed that low-risk group showed superior OS in ccRCC cohort (HR, 0.52, 95%CI, 0.37-0.75, P<0.001) and melanoma cohort (HR, 0.27, 95%CI, 0.12-0.62, P=0.001). CONCLUSIONS: Our study showed that the CCCP risk score is an independent biomarker that predicts the efficacy of ICIs in mUC patients. The patients with a low-risk score tend to have a better response to ICIs and a longer life time probably due to the immune-activated TME. Further studies are needed to validate the clinical utility of the seven-gene signature.

This study aimed to treat real wastewater from the desulfuration and denitration process in a petrochemical plant with high-strength nitrogen (TN≈200 mg/L, > 90% nitrate), sulfate (2.7%) and extremely low-strength organics (CODCr < 30 mg/L). Heterotrophic denitrification of multistage anoxic and oxic biofilm (MAOB) process in three tanks using facultative denitrifying bacteria inoculum was developed to simultaneously achieve desirable effluent nitrogen and organics at different hydraulic retention time (HRT) and carbon to nitrogen (C/N) mass ratios. The optimum condition was recommended as a C/N ratio of 1.5 and a HRT of A (24 h)/O (12-24 h) to achieve > 90% of nitrogen and organics removal as well as no significant variation of sulfate. The denitrifying biofilm in various tanks was dominant by Hyphomicrobium (8.9%-25.7%), Methylophaga (18.6%-25.8%) and Azoarcus (3.3%-19.6%), etc., containing > 20% aerobic denitrifiers. This explained that oxic zone in MAOB process also exhibited simultaneous nitrogen and organics removal.

Tetrabromobisphenol A (TBBPA) is a common brominated flame retardant that has a wide range of toxic effects on organisms. However, the mechanism of the toxic effects of TBBPA on the digestive system has rarely been studied. The purpose of this study was to investigate the mechanism of TBBPA toxicity on the gastric mucosa. In this study, TBBPA (mixed with corn oil) was administered by gavage at doses of 0 mg/kg (CG), 10 mg/kg and 20 mg/kg. The results showed that the levels of ROS, MDA and LPO were increased, and the activities of antioxidant enzymes were decreased. Large amounts of ROS activated the NF-κB pathway, leading to the development of an inflammatory response. The expression of BCL family and Caspase (Cas) family genes was increased, inducing apoptosis. The RIP3/MLKL pathway was activated, leading to cell necrosis. In summary, TBBPA can cause damage to the gastric mucosa through oxidative stress, leading to increased ROS activation of the NF-κB pathway. Treatment with the antioxidant NAC alleviated the damage to the gastric mucosa caused by TBBPA.

Obstructive sleep apnea-hypopnea syndrome (OSAHS) is a chronic and common sleep-breathing disease that could negatively influence lives of patients and cause serious concomitant diseases. Polysomnography(PSG) is the gold standard for diagnosing OSAHS, but it is expensive and requires overnight hospitalization. Snoring is a typical symptom of OSAHS. This study proposes an effective OSAHS screening method based on snoring sound analysis. Snores were labeled as OSAHS related snoring sounds and simple snoring sounds according to real-time PSG records. Three models were used, including acoustic features combined with XGBoost, Mel-spectrum combined with convolution neural network (CNN), and Mel-spectrum combined with residual neural network (ResNet). Further, the three models were fused by soft voting to detect these two types of snoring sounds. The subject's apnea-hypopnea index (AHI) was estimated according to these recognized snoring sounds. The accuracy and recall of the proposed fusion model achieved 83.44% and 85.27% respectively, and the predicted AHI has a Pearson correlation coefficient of 0.913 (R2 = 0.834, p < 0.001) with PSG. The results demonstrate the validity of predicting AHI based on analysis of snoring sound and show great potential for monitoring OSAHS at home.

Perovskite solar cells have rapidly developed as a promising technology for the next generation of low-cost photovoltaics, receiving enormous attention from researchers and industries. Compared to traditional semiconducting materials, metal halide perovskite exhibits outstanding tolerance to extrinsic ions. At a certain range of doping concentration, the interstitial occupancy of extrinsic ions provides appealing benefits to the perovskite films, contributing to higher performance and stability of the devices. This review summarizes the research progress of interstitial ions for metal halide perovskite, providing insights into the mechanism and identification of interstitial doping of extrinsic ions, covering the benefits of interstitial ions in regulating crystal growth, inhibiting ion migration, and reducing defect density. Finally, based on the latest progress and findings, further topics and directions of research on interstitial ions in metal halide perovskite are proposed to advance the understanding of interstitial ions in perovskite and promote the development of perovskite photovoltaic technology.

In order to promote the commercialization of perovskite solar cells, gas-quenching is considered to be a promising technique for perovskite film fabrication. However, when handling with methylammonium-free (MA-free) perovskites, it is often difficult to obtain high-quality perovskite films by gas-quenching. Herein, formate additives are employed to regulate the crystallization of MA-free perovskite, and improve the quality of perovskite films. Different additives of formamidine formate (FAFO) and potassium formate (KFO) is compared to investigate the role of formate groups in the crystallization of perovskite films prepared by gas-quenching. The FAFO additive facilitates the perovskite crystallization in (001) orientation whereas KFO favors for (111) orientation. The MA-free device with addition of FAFO demonstrate a champion power conversion efficiency of 20.94 %, compared to that of 20.14 % for KFO devices. In addition, FAFO device also exhibits improved thermal stability in ambient condition without encapsulation, extending the T80 lifetime by 18 times compared to the pristine device.

Potassium ion (K+) plays an important role in the maintenance of cellular biological process for human health. Thus, the detection of K+ is very important. Here, based on the interaction between thiamonomethinecyanine dye and G-quadruplex formation sequence (PW17), K+ detection spectrum was characterized by UV-Vis spectrometry. The single-stranded sequence of PW17 can fold into G-quadruplex in the presence of K+. PW17 can induce a dimer-to-monomer transition of the absorption spectrum of cyanine dyes. This method shows high specificity against some other alkali cations, even at high concentrations of Na+. Further, this detection strategy can realize the detection of K+ in tap water.

The strategic choice of state-owned enterprises (SOEs) is crucial to the sustainable development of China's economy. This paper explores the impact of mixed-ownership reform on the strategic choice of SOEs from the shareholder power and the board power. We find that the greater the diversity of mixed shareholders, the depth of mixed equity, the control of mixed equity, and the excess control of mixed equity, the higher the degree of mixed-ownership reform, and the more likely it is to promote SOEs to choose the prospector strategy. The mechanism test states that the impact of mixed-ownership reform on enterprise strategy is achieved through the balance effect between non-state-owned shareholders and state-owned controlling shareholders with the same power, and the synergy effect between different powers of non-state shareholders. Further research indicates that the mixed-ownership reform has a stronger driving effect on the prospector strategy in SOEs under strict external supervision, competitive industries, and local areas. This study clarifies the governance logic of non-state-owned shareholders on the strategic positioning of SOEs by dual control rights, and it provides empirical evidence for the formulation of enterprises' market-oriented strategic objectives.

Objective.Snoring is a typical symptom of Obstructive Sleep Apnea Hypopnea Syndrome (OSAHS). In this study, an effective OSAHS patient detection system based on snoring sounds is presented.Approach.The Gaussian mixture model (GMM) is proposed to explore the acoustic characteristics of snoring sounds throughout the whole night to classify simple snores and OSAHS patients respectively. A series of acoustic features of snoring sounds of are selected based on the Fisher ratio and learned by GMM. Leave-one-subject-out cross validation experiment based on 30 subjects is conducted to validation the proposed model. There are 6 simple snorers (4 male and 2 female) and 24 OSAHS patients (15 male and 9 female) investigated in this work. Results indicates that snoring sounds of simple snorers and OSAHS patients have different distribution characteristics.Main results.The proposed model achieves average accuracy and precision with values of 90.0% and 95.7% using selected features with a dimension of 100 respectively. The average prediction time of the proposed model is 0.134 ± 0.005 s.Significance.The promising results demonstrate the effectiveness and low computational cost of diagnosing OSAHS patients using snoring sounds at home.
